Rho‐kinase inhibition by fasudil modulates pre‐synaptic vesicle dynamics

چکیده

The Rho kinase (ROCK) signaling pathway is an attractive therapeutic target in neurodegeneration since it has been linked to the prevention of neuronal death and neurite regeneration. isoquinoline derivative fasudil a potent ROCK inhibitor, which already approved for chronic clinical treatment humans. However, effects treatments on function are still unknown. We analyzed here primary rat hippocampal cultures. Neurons were stimulated with 20 Hz field stimulation we investigated pre-synaptic mechanisms parameters regulating synaptic transmission after by super resolution emission depletion (STED) microscopy, live-cell fluorescence imaging, western blotting. Fasudil did not affect basic or amount several proteins, but altered dynamics vesicles. reduced proportion actively recycling vesicles, shortened vesicle lifetime, resulting overall reduction response upon stimulation. conclude that does alter structure, accelerates turnover, decreases number released This broadens known spectrum this drug, suggests new potential uses.

LRRK2 kinase activity modulates presynaptic vesicle release

Mutations in Leucine-rich repeat kinase 2 (LRRK2) are the single most common cause of inherited Parkinson ́s disease (PD). Little is known about its involvement in the pathogenesis of PD mainly due to the lack of knowledge about the physiological role of LRRK2. Our previous results suggest that LRRK2 acts as a scaffold within the presynaptic bouton and that it is involved in neurotransmitter rel...

Kainate modulates presynaptic GABA release from two vesicle pools.

Inhibitory control of local neuronal circuits is critical for prefrontal cortical functioning. Modulation of inhibitory circuits by several neuromodulators has been demonstrated, but the underlying mechanisms are unclear. Neuromodulator effects on synaptic vesicle recycling have received little attention. Controversy also exists whether different pools of synaptic vesicles underlie spontaneous ...

GABAB-Mediated Presynaptic Inhibition of Excitatory Transmission and Synaptic Vesicle Dynamics in Cultured Hippocampal Neurons

Local recycling of synaptic vesicle membrane at nerve terminals is necessary to maintain a readily releasable pool of transmitter. To what extent are the dynamics of vesicle recycling subject to modulation? We examined the influence of presynaptic GABA(B) receptors on vesicle dynamics at single synapses using optical imaging of FM1-43 in cultured rat hippocampal neurons. The kinetics of FM1-43 ...

G protein -subunits activated by serotonin mediate presynaptic inhibition by regulating vesicle fusion properties

Neurotransmitters are thought to be released as quanta, where synaptic vesicles deliver packets of neurotransmitter to the synaptic cleft by fusion with the plasma membrane. However, synaptic vesicles may undergo incomplete fusion. We provide evidence that G protein-coupled receptors inhibit release by causing such incomplete fusion. 5-hydroxytryptamine (5-HT) receptor signaling potently inhibi...

ROCK Inhibition by Fasudil Ameliorates Diabetes-Induced Microvascular Damage Short running title: Fasudil ameliorates diabetic microvasculopathy

OBJECTIVE: Leukocyte adhesion in retinal microvasuculature substantially contributes to diabetic retinopathy (DR). Involvement of the Rho/ROCK pathway in diabetic microvasculopathy and therapeutic potential of fasudil, a selective ROCK inhibitor, are investigated. RESEARCH DESIGN AND METHODS: Localization of RhoA/ROCK and Rho activity were examined in rats’ retinal tissues. Impact of intravitre...
